Usingcomputationalmodeling,wepredictthatthelimitedavailabilityofneurotransmitterresourcesincombinationwith thespontaneousreleaseofvesicleslimitsthemaximumdegreeofenhancementofsynaptictransmission.Thisgivesriseto anoptimaltuningthatdependsonthenumberofactivezones.Thereisstrongexperimentalevidencethatastrocytesthat enwrap synapses can modulate the probabilities of vesicle release through bidirectional signaling and hence regulate synaptic transmission. For low-fidelity hippocampal synapses, which typically have only one or two active zones, the predictedoptimalvalueslieclosetothosedeterminedbyexperimentallymeasuredastrocyticfeedback,suggestingthat astrocytesoptimizesynaptictransmissionofinformation.
