丹参对幼年大鼠应激性溃疡预防作用机制实验探究  【摘要】 目的:探讨丹参注射液对幼年大鼠应激性溃疡(SU)粘膜损伤预防作用及可能机制。方法:将36只幼年大鼠随机分成6组,即正常对照组、应激3 h组(Ⅰ组)、应激前腹腔注射生理盐水2 mL+应激3 h组(Ⅱ组)、应激前腹腔注射丹参2次+应激3 h组(Ⅲ组)、应激前腹腔注射丹参4次+应激3 h组(Ⅳ组)、应激前腹腔注射丹参6次+应激3 h组(Ⅴ组),采用束缚水浸法制作应激性溃疡动物模型,检测各组胃粘膜溃疡指数(UI)、胃粘膜血流量(GMBF)及胃组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)、肿瘤坏死因子(TNFα)含量变化。结果:(1)丹参可明显减轻损伤模型中的UI;(2)丹参能明显降低幼鼠胃组织和血清MDA含量,增加胃组织和血清SOD活性;(3)丹参可以降低幼鼠胃组织中TNFα的水平。Ⅲ、Ⅳ、Ⅴ组TNFα值均低于Ⅰ组,且依次递减,Ⅴ组与Ⅰ组相比差异有统计学意义(Plt;0.05)。结论:丹参对应激性溃疡具有预防作用,其机制与改善胃微循环、抗氧化清除自由基及通过对单核巨噬细胞分泌TNFα的调节等作用有关。 【关键词】 丹参;应激;胃粘膜;溃疡;自由基;肿瘤坏死因子 [Abstract] Objective: To investigate the prophylactic effect of Radix Salvia Miltiorrhiae Injection (RSM) on acute gastric mucosal injury and to explore possible mechanism in young rats.Methods: Thirtysix SD young rats were randomly divided into six groups. Different times of RSM (2 times, 4 times and 6 times per rat before gastric mucosal injury induced by water restraint stress) were used to treat the rats, then the change of gastric mucosal ulcer index (UI), pH of gastric juice, gastric mucosal blood flow (GMBF), the activity of SOD, the concentration of MDA and TNFα in gastric mucosa were measured, and a comparison was made among all the groups. Results:(1) RSM could remarkably decrease injury index and the concentration of MDA and TNFα in gastric mucosa; (2) RSM could significantly increase gastric mucosal blood flow (GMBF) and the activity of SOD; (3) RSM could markedly decrease the TNFα level of young rats in stomach tissue (Plt;0.05). Conclusions:RSM has a good protective effect on gastric stress ulcer caused by water immersion in young rats. The mechanism may be related to its increase of GMBF, the antioxidantive ability which can reduce injury induced by free radicals, and reduction of the levels of TNFα in gastric tissue.
