古桂遗传多样性与亲缘关系的ISSR分析  段一凡,王贤荣 ,梁丽丽,李雪霞,向其柏,赵正萍 (南京林业大学森林资源与环境学院,南京,210037) 摘要:利用ISSR分子标记,以云南木犀(Osmanthus yunnanensis (Franchet) P.S.Green)和柊树 (Osmanthus heterophyllus (G.Don. )P.S.Green )为对照种,对6个省市45份古桂花和2份野生桂花 进行了遗传多样性研究。结果表明:古桂花具有丰富的遗传多样性,选用的14条引物共扩增出148条 DNA条带,其中多态性条带131条,占88.51%;平均观察等位基因数、有效等位基因数、Nei’s遗传 多样性指数和Shannon多样性信息指数分别为1.8851、1.4106、0.2415和0.3688 ;聚类分析结果表明对 照种与桂花遗传距离较远,最早分开;古桂花具有很强的地理相关性,来自同一来源地或具有相似 颜色的个体往往聚在一起;四季桂与其他秋桂类个体遗传距离较远。 关键词:古桂;ISSR ;遗传多样性;聚类分析 中图分类号:S792.95;TQ920.1 Analysis of genetic diversity and relationships among ancient sweet osmanthus trees using ISSR markers DUAN Yi-fan,WANG Xian-rong ,LIANG Li-li,LI Xue-xia,QIANG Qi-bai,ZHAO Zheng-pin (College of Forest Resources and Environment, Nanjing Foresty University,Nanjing,210037) Abstract: Interspecific relationships of 45 ancient sweet osmanthus trees from 6 provinces and 2 wild species of sweet osmanthus were evaluated by using ISSR (inter simple sequence repeat) marker with Osmanthus yunnanensis (Franchet) P.S. Green and Osmanthus fordii Hemsl as contrast species. The results showed that ancient sweet osmanthus trees had abundant genetic diversity. Fourteen ISSR primers produced a total of 148 bands, of which 131 were polymorphic. The percentage of polymorphic bands was 88.51%. The mean observed number of alleles, effective number of alleles, Nei’s genetic distance and Shannon index were 1.8851, 1.4106, 0.2415 and 0.3688 respectively. The cluster analysis showed that the genetic distance between contrast species and sweet osmanthus was far.
