大黄鱼复方中草药氧化氢酶基因表达.doc

PAGE 　　 摘 要 过氧化氢酶（CAT）是生物体中的一种抗氧化酶，本文以我国重要的海水养殖鱼类大黄鱼为研究对象,探究饲料中添加复方中草药对大黄鱼过氧化氢酶基因表达的影响。通过在大黄鱼饲料中添加不同比例中草药复方（太子参、巴戟、当归、枸杞、茯苓、白术等），饲喂30天后，以大黄鱼的肾脏组织为实验材料，通过实时荧光定量PCR技术，分析饲喂中草药后CAT基因的相对表达变化。结果显示：与对照组相比，各实验组中该基因的表达均出现了不同程度的下降（E2组除外），E5组的表达量下降最多（72%），E1、E3、E4组的CAT基因表达量的下降程度差不多，均在40%-50%之间，E2组的表达量比对照组升高41%，但是各组间均无显著性差异（P大于0.05）。本研究结果为中草药作为饵料添加剂在大黄鱼饲料中的应用提供了基础数据，也为中草药作为鱼类免疫增强剂提供了前期研究基础。 关键词：大黄鱼；复方中草药；过氧化氢酶；基因表达 Abstract In?this?paper,?yellow?croaker,?an?important?mariculture?fish?in?China,?was?taken?as?the?research?object.?In?order?to?explore?the?effect?of?compound?Chinese?herbal?medicine?on?the?catalase?(CAT)?gene?expression?of?yellow?croaker, this experiment analyzed the relative expression changes of cat gene after feeding Chinese herbal medicine by adding different proportions of compound Chinese herbal medicine (Pseudostellaria heterophylla, Morinda officinalis, angelica sinensis, medlar, poria cocos, atractylodes macrocephala, etc.) into the feed of yellow croaker and taking kidney tissue of yellow croaker as experimental material after 30 days of feeding. The results showed that Compared with the control group, the expression of this gene in all the experimental groups decreased to different degrees (except the E2 group), and the expression of E5 group decreased the most (72%). the expression of CAT gene in E1, E3 and E4 groups decreased to the same degree, ranging from 40% to 50%.
